A twelve-year-old Labrador mix, 68 pounds, comes in because her owner read about NAD+ on a wellness forum at 11 p.m. and wants to split her own capsule with the dog “just to try it.” That’s the scenario I see now more than I did even two years ago, and it’s worth walking through carefully rather than answering with a reflexive no.

Why the same molecule doesn’t mean the same product

NAD+ (nicotinamide adenine dinucleotide) is a coenzyme every cell needs for energy metabolism and DNA repair, and it does decline with age in most mammals, dogs included. That part isn’t controversial. What’s controversial — or at least unsettled — is whether the precursor compounds sold to boost it, mainly nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN), belong in a dog’s bowl in the same form and dose built for a 150-pound adult human.

Human NAD+ capsules are formulated, tested for tolerability, and dosed against human body weight and human safety data. A 2017 randomized, placebo-controlled trial of nicotinamide riboside with pterostilbene in humans reported the combination increased NAD+ levels safely and sustainably over eight weeks, according to a study published in PMC. That’s real, useful evidence — for humans. It says nothing about a 20-pound terrier’s liver enzymes, and the FDA’s own labeling framework draws the line explicitly: guidance from FDA treats animal-intended products as a distinct regulatory category from human dietary supplements, and NASC/FDA-CVM conventions don’t even permit instructions for repurposing a human label onto a pet.

Where the dog-specific evidence actually stands

This is the part I want to be honest about, because it’s thinner than the marketing copy suggests but not nothing. A randomized, controlled trial run at NC State tested a combined NAD+ precursor and senolytic compound in seventy senior dogs with mild to moderate cognitive impairment, split across placebo, low-dose, and full-dose groups. Fifty-nine dogs completed the three-month primary endpoint. Owner-reported cognitive scores improved significantly more in the full-dose group than placebo, according to the trial published in Scientific Reports. That’s a dog-specific, dosed, controlled study — genuinely stronger evidence than most supplement categories can claim. It’s also one trial, six months, one formulation, and it doesn’t tell us anything about a human capsule split in half.

That gap between “NAD+ decline is real in dogs” and “this specific human product at this specific dose is safe for dogs” is where I ask owners to slow down. Related antioxidants marketed alongside NAD+ precursors carry the same caution. Quercetin’s cellular effects have mostly been characterized outside the dog: one lab study found quercetin, along with NAC and ascorbic acid, “did not affect the feeding behavior” of the model organism tested, according to research indexed under DOI Pharmaceuticals (DOI.3390/ph19040525): 10.3390/ph19040525 — informative about mechanism, not a dosing guide. Resveratrol data is similarly early-stage; one embryonic-exposure study found a specific concentration “accelerated pupation and adult emergence but shortened adult lifespan” in the model species studied, per research at DOI Phytomedicine (DOI.1016/j.phymed.2026.158531): 10.1016/j.phymed.2026.158531. Neither of those findings translates cleanly to a senior dog’s bowl, and I don’t pretend they do.

The tradeoff owners run into when they try it anyway

I had a case that still shapes how I answer this. A nine-year-old spayed female Cavalier, 17 pounds, whose owner had been giving her a quarter of a human NR capsule for three weeks because a friend swore by it for her own dog. No acute crisis, no ER visit — but no way to know the actual milligram dose the dog was getting either, because human capsules aren’t scored or weight-banded for a 17-pound frame. By day 21 the owner couldn’t tell me if the dog was getting too little to matter or enough to matter, and neither could I. That’s the real failure mode with human NAD+ formulas in dogs: not usually acute toxicity, but an unmeasurable, unverifiable dose that makes the whole exercise unfalsifiable. You can’t titrate what you can’t quantify.

Here’s the wrinkle I didn’t expect early in my integrative practice: breed size doesn’t just change the math, it can change the recommendation entirely. A 6-pound Chihuahua and a 90-pound Rottweiler metabolize B-vitamin-family compounds like NR differently in absolute terms, and a human dose split “in half for the little dog” can still land wildly high on a per-pound basis for the small one while landing low for the large one — the opposite of what most owners assume. Where I land, and the counterargument I take seriously

My position: don’t give a dog a human NAD+ formula, and reach for a dog-dosed nicotinamide riboside chew instead if you and your veterinarian decide the category is worth trying for a senior dog’s cognition or activity level. The strongest counterargument is a fair one — NR and NAD+ biology is conserved across mammals, the human safety data is reassuring at the doses studied, and a careful owner splitting a capsule isn’t reckless in the way giving a dog ibuprofen would be. I don’t dismiss that. But “conserved biology” isn’t the same as “validated dose,” and the one trial we do have in dogs used a purpose-built, dog-dosed formulation, not a repurposed human capsule. I’d rather defend a position built on the dog-specific RCT than extrapolate confidently from human pharmacokinetics I haven’t seen replicated at canine body weights.
